Unit Definition: One unit (U) corresponds to the enzyme quantity required to hydrolyze cellulose substrate into reducing sugars equivalent to 1 mg of glucose per hour under standardized conditions (pH 4.8, 50°C).
Mechanism: Cellulase is a multi-enzyme system comprising:
- Endoglucanases (EGs): Randomly cleave internal β-1,4-glycosidic bonds in cellulose chains.
- Exoglucanases/Cellobiohydrolases (CBHs): Processively hydrolyze cellulose from polymer ends, releasing cellobiose.
- β-Glucosidases (BGs): Convert cellobiose into glucose by breaking β-1,4 bonds.
